Oligo Array spotted oligonucleotide, UTSW Human 22k Oligo spotted array, Construction and Preparation of Human Oligo 22K Spotted Microarrays by UTSW: Operon Human Genome Oligo Set (Version 2) (70mers) representing 21,329 human genes in 150mM Sodium Phosphate (pH8.3) plus 1xSSC at 30 µM were spotted singly onto polymer-coated glass slides (Full Moon Biosystems) using BioRobotics MicroGrid II spotter (Genomic Solutions) fitted with 4x12 Microspot 2500 quill pins at 20-23°C and 60-65% RH. Spot are arranged with 22x22 spots per block (48 total blocks) and spot centers are 200 µm apart. Printed slides were subjected to a UV-cross-linking at a dose of 650mJ cm2 for probe immobilization. Slides with immobilized probes were then stored in slide holders and placed in a desiccator at room temperature.

Spotting was performed in duplicates (replicate arrays) with 4x6 SMP3 pins (Telechem) at 18-20 °C and 40-50% RH on epoxysilane coated slides (Quantifoil) using 40 µM oligos in FBNC spotting buffer (G. Wrobel et al., Nucleic Acids Res 31 e67, 2003). Spot centers were 129 µm apart. DNA adhesion was accomplished by 1 h incubation at 60 °C followed by 240 mJ/cm2 UV irradiation at 254 nm.
