Tags were blunted with T4 polymerase and ligated to generate ditags. Ditags were PCR amplified and digested with NlaIII. Restriction fragments were separated on a 12% polyacrylamide gel and the band containing the ditags was isolated from the gel. The purified inserts were concatemerized and ligated into pZero and transformed into EDH5? electromax cells.
