(2011) Transcriptome analysis of the desert locust central nervous system: production and annotation of a Schistocerca gregaria EST database. PLoS One 6: e17274.]. The microarray platform contains probes corresponding to all these S. gregaria EST sequences and to all available GenBank S. gregaria transcript sequences. The migratory locust (Locusta migratoria) is a species related to the desert locust for which EST sequence information is also available [Ma ZY, Yu J, Kang L (2006) LocustDB: a relational database for the transcriptome and biology of the migratory locust (Locusta migratoria). BMC Genomics 7: 11.]. Therefore, the S. gregaria transcript sequences included in the microarray design were complemented with all L. migratoria EST sequences having no blastn orthologue (blastn hits producing E < 1E-10 were considered orthologs) in the S. gregaria set. The locust microarrays were designed by means of the eArray system (Agilent Technologies). Probes were designed according to the following criteria: (i) probe length: 60 nucleotides, (ii) two probes per target sequence (best probe methodology) and (iii) probe orientation: sense. Other parameters were set at default settings. In total 20,755 unique locust transcript sequences (12,783 S. gregaria and 7,972 L. migratoria sequences) were represented on the arrays. (EST sequences are publically available in GenBank.) |
