GeneChip spotted DNA/cDNA, Eucalyptus xylem microarray (NCSU), The cDNA microarray comprises 2608 ESTs, printed in duplicate. Each cDNA has been printed by Pin 1 and 4, or by Pin 2 and 3. 555 ESTs represent the UniGene set derived from the sequencing of a cDNA library from Eucalyptus grandis differentiating xylem generated by the Forest Biotechnology Group at NCSU (egx prefix). The remaining 2053 ESTs are from four libraries from Eucalyptus grandis leaf and root tissue and two libraries from Eucalyptus tereticornis flower tissues, kindly made available by DuPont (Scott Tingey, DuPont Crop Genetics, Newark, DE). These ESTs were selected from a collection of approximately 13,000 sequences, based on their putative function in the biogenesis of cell walls, biogenesis of the cytoskeleton, biosynthesis of phenylpropanoids, protein targeting, c-compound and carbohydrate metabolism, cell growth, signal transduction, protein transport, energy metabolism and other functions potentially associated to wood formation. The cDNA clones were amplified by PCR, purified (Millipore 96-well Multiscreen filter plates) and the quality of the amplification evaluated using agarose gels. Preparations with multiple bands and low quality amplified cDNAs were eliminated and repeated. After dilution in 50% DMSO, cDNAs were printed in duplicate on aminosilane-coated glass slides (Corning) using an Affymetrix 417 Spotter. Keywords = Eucalyptus xylem
