Arabidopsis ecotype Nossen wild-type plants were grown on solidified MS medium. Total RNAs were prepared from irradiated / non-irradiated seedlings using an RNeasy Mini Kit (Qiagen, Hilden, Germany). 400 ng of each RNA sample was reverse transcribed, labeled with Cy3, Cy5, and purified with an RNeasy Mini Kit (Qiagen). One microgram each of Cy3- and Cy5-labeled cRNA was added to 500 ul of the hybridization buffer and the mixtures were applied to microarrays under cover slips (Arabidopsis 2 Oligo Microarray, Agilent). |
