Each aliquot was ligated to one of the two annealed linker pairs. After extensive washing tags were created by digestion with BsmFI (tagging enzyme). Tags were blunted with T4 polymerase and ligated to generate ditags. Ditags were PCR amplified and digested with NlaIII. Restriction fragments were separated on a 12% polyacrylamide gel and the band containing the ditags was isolated from the gel. The purified inserts were concatemerized and ligated into pZero and transformed into EDH5? electromax cells. |
