The GeneChip E. coli Antisense Genome Array can be used for examining expression of all known E. coli genes and discovering novel, uncharacterized intergenic transcripts. The array contains probe sets to detect the antisense strand of more than 4,200 known open reading frames. In addition, over 1,350 intergenic sequences can be interrogated in both directions permitting functional characterization of potentially important shorter expressed sequences. cDNA Array spotted DNA/cDNA, QtsA cDNA clones, We determined 512 full-insert sequences of cDNAs derived form cynomolgus monkey testis. An aliquot of the same DNA preparation used in the 5’-end-sequencing reactions provided material for the PCRs. Inserts were amplified by PCR using 5’-CTTCTGCTCTAAAAGCTGCG-3’ as a forward primer and 5’-CGACCTGCAGCTCGAGCACA-3’ as a reverse primer, in a volume of 100 µl. Successful amplification was confirmed by agarose gel electrophoresis. When the first PCR failed to amplify enough products, the first PCR products were amplified again. Samples of approximately 300 µg /ml DNA in 2 x Solution-T reagent (Takarabio) were printed on duplicate glass-slides with a GMS 417 arrayer (Genetic MicroSystems).
